Serodiagnosis of hydatidosis in livestock by the indirect hemagglutination test (lha) and the enzyme-linked immunosorbent assay (elsa)
Abstract
In an attempt to improve the serodiagnosis of hydatid disease in livestock, an antigen designated as antigen "880" was purified from bovine hydatid cyst fluid (HCF) and used in the IHA and ELISA tests. The antigen was concentrated and partially purified by precipitation with phosphotungstic acid and magnesium chloride solutions, and finally purified by affinity chromatography using an an¬tiserum specific for this antigen. Antigen "880" was shown to.be part of the antigen structure of 5 out of 14 other parasites. The five parasites were: Taenia saginata, Cysticercus tenuicollis, Moniezia expense, Avitellina centripunctata and Oesophagostomum radiatum.
Although antigen "880" may be a lipoprotein common to several parasites our results showed a sensitivity of about 93% and a specificity of 99% in the IHA test. In the ELISA, the sensitivity was 91 % and the specificity was 100%. These tests were carried out on sera from goats, sheep and cattle, naturally infected with hydatid disease.
